چکیده

As you can see from their comments, while referee #1 is rather negative towards your study, referees #2 and #3 are very supportive of its publication and agree on the high potential interest of your findings. In general, although they believe that the evidence presented properly supports your conclusions, some technical concerns have arisen with which you will have to deal before your manuscript is ready for publication. As referee reports are quite explicit I will not repeat their arguments here, but I would like to draw your attention to points 1 and 6 from referee #2 -regarding Lats phosphorylation site and the role of NF2 respectivelyand major point 1 from referee #3 -on the effects of PKA on Lats activityas important concerns that need to be adequately addressed.

cAMP/PKA signalling reinforces the LATS-YAP pathway to fully suppress YAP in response to actin cytoskeletal changes.

Actin cytoskeletal damage induces inactivation of the oncoprotein YAP (Yes-associated protein). It is known that the serine/threonine kinase LATS (large tumour suppressor) inactivates YAP by phosphorylating its Ser127 and Ser381 residues. However, the events downstream of actin cytoskeletal changes that are involved in the regulation of the LATS-YAP pathway and the mechanism by which LATS diffe...

Protein kinase A activates the Hippo pathway to modulate cell proliferation and differentiation.

The Hippo tumor suppressor pathway plays an important role in tissue homeostasis that ensures development of functional organs at proper size. The YAP transcription coactivator is a major effector of the Hippo pathway and is phosphorylated and inactivated by the Hippo pathway kinases Lats1/2. It has recently been shown that YAP activity is regulated by G-protein-coupled receptor signaling. Here...

Arhgef7 promotes activation of the Hippo pathway core kinase Lats.

The Hippo pathway regulates tissue growth and organ size, and inactivation contributes to cancer. Signals flow through Mst/Lats kinases, which phosphorylate and promote cytoplasmic localization of the transcriptional regulators Yap and Taz to inhibit transcription. Here, we identify the multidomain-containing guanine nucleotide exchange factor (GEF) Arhgef7, or βPix, as a positive Hippo pathway...

Angiomotins link F-actin architecture to Hippo pathway signaling

The Hippo pathway regulates the transcriptional coactivator YAP to control cell proliferation, organ size, and stem cell maintenance. Multiple factors, such as substrate stiffness, cell density, and G protein-coupled receptor signaling, regulate YAP through their effects on the F-actin cytoskeleton, although the mechanism is not known. Here we show that angiomotin proteins (AMOT130, AMOTL1, and...

Hippo pathway regulation by cell morphology and stress fibers.

The Hippo signaling pathway plays an important role in regulation of cell proliferation. Cell density regulates the Hippo pathway in cultured cells; however, the mechanism by which cells detect density remains unclear. In this study, we demonstrated that changes in cell morphology are a key factor. Morphological manipulation of single cells without cell-cell contact resulted in flat spread or r...
